海藻酸钠浓度对新型神经组织工程支架结构与性能的影响Effect of Sodium Alginate Concentration on the Structure and Properties of a New Scaffold for Nerve Tissue Engineering

摘要(Abstract):
探讨海藻酸钠溶液的浓度变化对藻酸盐水凝胶新型组织工程支架结构与性能的影响.在氯化钙溶液浓度相同的条件下,改变海藻酸钠溶液的浓度,制备出不同原料配比的藻酸盐水凝胶,冷冻干燥后于光学显微镜下观察其微观结构变化,测定水凝胶的含水率与凝胶质量分数;将无菌的海藻酸钠溶液及间充质干细胞混合后,与氯化钙溶液反应得到包载干细胞的藻酸盐水凝胶,倒置荧光显微镜下观察水凝胶所释放出细胞的存活情况.在氯化钙浓度一定的条件下,随着海藻酸钠浓度的减小,水凝胶的空间网络结构由致密变得疏松,更易于细胞的生存与释放.实验表明,海藻酸钠水凝胶作为组织工程支架,用于神经损伤后修复具有较好的应用前景.
